What is a monoclonal antibody?
A monoclonal antibody is a specific antibody usually produced by hybridoma technology. A hybridoma is a fusion of a single antibody producing murine B cell with a myeloma cell to immortalize the antibody producing cell line. A monoclonal antibody recognizes a single epitope of its target protein, making it a very useful tool in research, diagnostics, and therapeutics. To produce a monoclonal antibody, a single hybridoma that produces the specific antibody of interest is isolated and expanded in culture. The antibody is then purified for use. Monoclonal antibody production can also be done by in vivo methods, which produces an ascites. The in vivo method involves injection of hybridoma cells into the peritoneum of mice where hybridoma cells grow and secrete the antibody of interest. Due to inflammation, ascites fluid is produced and collects in the peritoneum. The ascites fluid contains the antibody produced by the hybridoma cells as well as many other proteins, including background antibodies produced by the mice and albumin.
Antibody concentrations in ascites are typically high, but an accurate concentration of this antibody cannot be readily determined. Ascites is a complex solution containing multiple proteins, background antibodies, as well as the antibody of interest.
Monoclonal antibodies may be purified by various methods. The most common methods are protein A or protein G affinity chromatography. Most sub-classes of antibody have a high affinity for protein A and protein G, thus the antibody binds to these proteins on a resin and extraneous proteins can be washed away. The purified antibody can then be eluted off the resin.
Alternatively, antibodies can be affinity purified utilizing their target immunogen. This method can be expensive and relies on immunogen availability.
NeoClone typically utilizes protein A for purifying antibodies produced by in vitro technology. On occasion, when antibodies need to be purified from ascites, protein G is used.

In general, the more conserved proteins are between species, the more likely the antibody will cross react. If the epitope is known, an alignment can determine if the epitope sequence is conserved in a similar region of the protein.
